The Ready-to-Assay GPR54/KiSS1 Receptor Frozen Cells are a simple, thaw-and-assay cell format for use in studying drug candidates (small molecules or biologics) targeting GPCRs. This assay assesses ligand based activation of KISS1R GPCR activity via detection of increasing levels of intracellular calcium. The included cell line overexpresses KISS1R alongside endogenous high levels of a promiscuous G-protein, coupling the receptor to the calcium signaling pathway. Upon activation of KISS1R, the resulting calcium activity can be fluorescently assayed. Ready-to-assay kits consist of two vials of the stably modified clonal cell line in a convenient thaw-and-use format. The PathHunter FPRL1 cell line is a stable, engineered cell line for use in studying drug candidates (small molecules or biologics) targeting GPCRs. This cell line enables assessment of ligand (e.g. formylpeptides) based activation of FPRL1 GPCR activity via detection of β-arrestin recruitment. The included cell line overexpresses ProLink-tagged FPRL1 and EA-tagged β-Arrestin-2. Activation of FPRL1 stimulates the recruitment of β-Arrestin-2 and produces EFC signal. Ki-67 antigen is a nuclear, non-histone protein that is present in all stages of the cell cycle except G0. This characteristic makes Ki-67 an excellent marker for proliferating cells and is commonly used as one of the prognostic factors in cancer studies. Assessment of Ki-67 expression in renal and ureter tumors shows a correlation between tumor proliferation and disease progression, thus making it possible to differentiate high-risk patients.
